Amygen Worthington's tested cell isolation systems:
The Cell Dissociation Systems currently offered by Worthington are kits containing enzymes and other required reagents to perform tissue dissociation without the need to purchase individual packages of one or more enzymes and pre-test various batches of a certainsome enzymes.Some are designed for use with specific organizations, and one of the suites is a general program development system.Below are instructions for each current kit.

Worthington Hepatocyte Isolation System:
Most traditional methods published for the isolation of hepatocytes use crude and partially purified enzyme preparations, including various types of collagenases and other proteases.More recently, better-characterized collagenase preparations, such as Worthington types 1 and 4 (CLS-1, 4) have provided better results.All crude collagenase preparations may contain batch-variable contaminating proteases, esterases, and other enzymes, requiring researchers to pre-screen several batches of enzymes and/or continually modify separation parameters and protocols.
The Worthington Hepatocyte Isolation System was developed to provide researchers with a reliable, convenient and consistent system for the isolation of hepatocytes.By using the pre-optimized enzyme combinations included in this kit, you can minimize batch-to-batch variability and improve the quality of isolated hepatocytes.Additionally, Worthington conducts use testing of each batch by isolating hepatocytes from adult rats to ensure live cell performance, reliability and consistent yield.
The Emtechy Worthington Neonatal Cardiomyocyte Isolation System (NCIS):
The Worthington Neonatal Cardiomyocyte Isolation System was launched to provide researchers with a reliable, convenient and consistent cell isolation system.Batch-to-batch variability can be minimized by using purified rather than crude enzyme preparations.Additionally, Worthington tested the kit for use by isolating cardiomyocytes from neonatal rat hearts to ensure live cell performance, reliability, and consistent yield.
The kit was developed in collaboration with Dr. Ronald MacGregor of the University of Kansas Medical Center.This method is based on the method described by Toraason et al.(1988) incubate minced tissue with trypsin overnight at low temperature.As pointed out by Toraason, this step reduces the time required to harvest cells compared to the time involved in continuous incubation in warm trypsin or collagenase.
Ai Meiji Worthington Papain Dissociation System (PDS):
The Worthington Papain Dissociation System is a suite of reagents designed for use in the tissue dissociation method of Huettner and Baughman.The materials are designed for convenience and simplicity, and are useful to occasional users as well as more experienced and frequent users.Each batch is tested for tissue dissociation performance and provides freshly prepared enzyme solutions for each dissociation.
Reagents are stable at ambient temperature for the expected time during normal shipping, but packaging should be refrigerated upon arrival and stored at 4-8°C for up to four months prior to use.
Amigen Worthington Cell Isolation Optimization System (CIT):
Worthington Biochemical Corporation provides a complete solution of enzymes most commonly used in tissue dissociation and cell isolation.The "Cell Isolation Optimization System" includes instructions, references, and strategies for the handling, use, and optimization of enzymatic cell isolation methods to achieve maximum yield of viable cells.
The System contains all of the enzymes commonly used in tissue dissociation and cell isolation procedures, as well as cell isolation guidelines detailing commonly used tissue types, the mode of action of each enzyme, tissue culture techniques, and guidelines for protocol optimization.In addition, this guide lists hundreds of cell- and tissue-specific isolation references for getting started with enzymatic cell isolation.
Note: Worthington's tissue dissociation enzymes are used interchangeably for most of the preparations cited.
